For co-transfection prepare at least 10mg of highly purified plasmid DNA. Spodoptera frugiperda cells are sensitive to some contaminants found in crude plasmid preparations, which cannot be removed by phenol/chloroform extraction or ethanol precipitation.

It is a known, that the usage of plasmid expressing reporter gene driven by strong promoter causes problems when one wants to compare different cell lines. This method allowes you to measure directly the amount of the plasmid which entered the cells and therefore provides a reliable reference.

Transfection with Liposomes

Transfection of mammalian cell lines is usually accomplished with calcium phosphate or liposomes. Transfection by liposomes is rapid and has high efficiency, but is more expensive. The procedure is very simple in essence, but there are a large number of parameters which can be tweaked, including ratio of DNA to liposomes, amount of DNA per well, time of incubation etc.
