optimal SDS separation protein - (Sep/09/2005 )

Hi...

I just want to know how to well separate 60 and 62 KDa proteins, on SDS Page gel. And what is the right % to use?

thanks...

10% acrylamide. But it might help if you play a littlebit with the ratio acryl/bisacryl.

hi
i would try a 15% acry gel.

I agree that a high percentage is the best idea, but if you don't want to always have to guess what gel percentage to use for a given western, I've just got into the habit of always pouring gradient gels.

a 4-20% gradient gel will always have the desired seperation in both low and high molecular weights. It has a low gel percentage at the top to seperate large proteins and a higher percentage towards the bottom to seperate smaller proteins. They sell plastic gradient makers relatively inexpensively and aren't difficult to use.
