PQE vector - Getting protein in its soluble form (Sep/06/2005 )

I have cloned a protein ( N-terminus) in PQE vector and I want to get its soluble form to study its biological activities and its regulation pathway. I tried almost all possibilities to solubalise it but i was not able to get it in its native form and denatured form is not what i want sad.gif sad.gif sad.gif . Please give me some tips.

does this vector express a fusion protein (6His tag, Thioredoxin, etc)?

how well is the protein expressing?

ya it does. and the problem is it forms inclusion bodies, i think overexpressed.

when you say you've tried almost all possibilites do you mean growth and expression temperatures, IPTG concentrations, different cracking buffers?

if those don't work it all it's possible that the tag is interferring with the folding of the protein.....
