Hi,
I am extracting a protein from plant system, from a fruit that contains good amount of pectic material. The protein has good activity in crude extract for as many days , but looses activity upon subjecting to any type of column chromatograhy. Also I am not able to separate the pectic substances from my protein .Kindly help me by giving sugestions how to circumvent the problem.
Thank-you.

Check you have no Ca in your extraction buffer or use EDTA. Use pH above 8.5 if possible.
