separate proteins from sucrose - (Apr/21/2005 )

Hi! I want to separate my plasma membrane preparation from a sucrose solution (45%), because it is interfering with the running on SDS-PAGE.
I tried to precipitate the proteins with perchloric acid, but I think sucrose precipitated with my membranes, as the gel didnĀ“t run well. With cold acetone sucrose also precipitates.

Precipiate out the protein with ammonium sulphate. Otherwise, you can also pass the solution through a desalting gelfiltration column.

Since you already did the acetone wash, I would suggest to perform dialysis after the acetone ppt to get red of sucrose and any existing salts from your sample prep. Good Luck! biggrin.gif
