DNA precipitation EtOH with Na OAc or Ammonium OAc - (Nov/12/2008 )
Hi everyone
I really want to optimize DNA precip with EtOH and either Na or Ammonium OAc (depending on my needs). I have all my protocols ready and have read as much as I can about the two however, I know that sometimes when it comes to reagents one manufacturer might be better than anothers due to their synthesis, quality control, etc. So my question is....... if anyone out there who is getting great results with either Na OAc or Ammonium OAc precip....could you let me know what company you are using for that particular reagent

Ammonium is better than sodium for smaller fragments. I just use Sigma ammonium OAc. I don't think the process is so troublesome as to need a specific manufacturer.

The source of the chemicals should matter little. NaOAc is often chosen for precipitation since ammonium ions inhibit the enzyme T4 polynucleotide kinase. The sodium salt requires no thought on what the downstream use of the DNA will be.
