phrGFP-1 - searching for vector sequence - (Oct/31/2008 )
Hello
I have a big problem. I want to use a phrGFP-1 vector from Stratagene
http://www.stratagene.com/vectors/maps/pdf/phrGFP_1.pdf
but as You can see on the page the company show only a part of the vector sequence: MCS 1 and MCS 2. My problem is that i want to clone my DNA fragment into MCS 1 but the second cloning place is 34nt downstream from the kozak sequence so I need to know the part of the hrGFP coding sequence. Stratagene can giv me a entire vector sequence but only when i buy this vector and this is very stupid in my opinion.
So my question is that maby one of You used this vector and can give me a sequence? I will be very very gratefull because i cant use other vector.

Why don't you find the hrGFP sequence on GenBank: this is the accession number: EU048698. Remember to reverse complement this sequence before use ^^.
That is the stupidest thing i've heard in a while. Are they trying to sell you the vector or not? Amazing.

I don't have the full sequence, but I do have a restriction map for the entire vector if that would be any help.
I have the restriction map but thanks anyway:)
Someone who has it should sequence it and deposit the sequence in Genbank.
