Yeast Transformation Problems - (Oct/07/2008 )

Hi All,

I am trying to transform the yeast strain S.cerevisae AH109. It is for a Y2H screen. I want to transform my baits and control vectors in but have not had any success. I have remade my reagents, media etc on several occasions but have gotten no colonies at all. I have tried my three control vectors and six baits but have had no success with any. I have also tried my Y187 reporter strain (both were part of the clontech yeast 2 hybrid matchmaker 3 kit). I have retaken my strains from stock twice to ensure fresh growth. Does anyone have any suggestions??
