Transformation - thanks for help (Aug/10/2008 )

Dear Bioforumers:
Iam trying to clone scfv these days,actually my reagents are quite old.I did transformation yesterday and I got only white colonies and no blue ones.
should I do plasmid miniprep or suspect this result.

yes its always better to do the miniprep and check for the presence of DNA...considering that thr could be self ligated plasmid too.. huh.gif

How can I know that my IPTG and XGal working or not

Do you have any of your vector left? Surely all you need to do is a vector ligation control on this sample and they should all be blue.

I always used to run a vector alone control in my cloning for this very reason (and sometimes an insert alone to check for any carry over of donor vector).

Thanks I did minipreps and RePCr pf plasmids the results were excellent
