6XHistaq proteins - (Jul/27/2008 )
Dear All
would you please tell me about 6XHistaq which are combined in N-terminal of Fusion protein (6KD+35KD) Expressed in pQE30?
is it conflict with folding of fusion protein?
is it recommended for cleavage of this marker after purification of protein?
if yes, which protocol do you recommend?
very thanks

it is not necessary to cleave off the histag after purification unless if you want to do MS. But even that, it is not necessary to cleave.

You should always cleave an affinity tag whenever possible to avoid the obvious question by a reviewer/examiner that the his tag may be interfering/modifying with the protein function.
Why don't you read the Qiagen manual for your vector and that will tell you how to cleave the his tag. It must be available on their website.
