NP40 can be used for lysis nulear membrane? - (Jul/15/2008 )

hi everybody, NP40 can be used for lysis nulear membrane except SDS ? And about concentration, can I use 1% ?

Yes, in my experience. If you freeze-thaw/sonicate/use high salt/incubate long etc, you can get it to lyse even at .5%.

So who can tell me the difference of NP40 between cell lysis buffer and nuclear lysis buffer????

I have this from Invitrogen:

NP40 Lysis Buffer –
150mM NaCl
1% NP-40
50mM Tris pH7.5
1.5mM EDTA
2 ug/ml Aprotonin
2μg/ml Leupeptin
1mM PMSF
H20


RIPA Lysis Buffer –
150mM NaCl
1% NP-40
0.1%SDS
50mM Tris pH 7.5
2 ug/ml Aprotonin
2μg/ml Leupeptin
1mM PMSF
1.5 mM EDTA
H20
