protein recovery problems in analytical size exclusion chromatogrphay - or how do i have a more sensitive method? (Jun/26/2008 )
hi all..
i have protein recovery problems in analytical size exclusion chromatography? what are the chanegs i can make to the existing method/ mobile phase? its an ammonium bicarbonate buffer, pH 7. and why? thanks.
or how do i have a more sensitive method in such a case?

maybe the inclusion of 10% EtOH is to reduce nonspecific absorption?

I've never heard of using EtOH to reduce nonspecific absorption - it was always neutral salt like NaCl (0.1-0.5 M).

High salt and maybe a touch of NP40 (0.02%). Are you may be planning to lyophilize afterwards? Is that the reason of using ammonium carbonate?
