cloning into the pBAD-GFPuv - (Jun/17/2008 )
Haloo every one, has anyone experiences with cloning in to the pBAD-GFPuv I need to clone gene of my interest in EcoRI anKpnI site, which are one bp behind GFP ORF. I would like to create fusion protein. But this GFP ORF ends with TAA stop codon and I think translation will end there and no fusion complete. ORF include all suitable RS so it is hardlly possible to design primers with RS and subclone it to another vector. Partial digestion suppose to by way but I dont like it
