problem on membrane preparation - (Sep/07/2004 )

I followed the protocol in our lab to prepare cell membrane from mice tissue. But I got membrane much less than what they did before. We discussed over but it seems the only reason will be the insufficient homogenization. However, I really think I chopped the tissue small enough, so I'm wondering what will be the key to reduce the yield of the membrane because I really don't understand very well about the protocol: why we can get membrane at this speed of centrifugation?

Is there anyone have a good idea of this?

In our lab we have experience in tissue homogenization and membrane purification, but given that the different protocols are slightly different, would be fine that you indicate the protocol that you apply for mice.
