ِpeptide sequencing - (May/01/2008 )

Hi everyone:

1- I have identified the amino acid sequence of complement factor B using bioinformatics tools. my question is how can I confirm this sequence experimentally? Can any one help?

2- What type of experiment should I do to obtain the physical properties of CFB?

q1: is it really an identification or only a prediction? you need the genuine nucleotide or amino acid sequence

q2: Mw (gel filtration), pI (chromato focussing, 2D GE), STOKEĀ“s radius (sucrose density gradient centrifugation), sedimentation coefficient (sucrose density gradient centrifugation), friction ratio (sucrose density gradient centrifugation)

1) If you can purify it, either Edman sequencing or you could try MS/MS
2) Also try CD for general structural data
