in tris-glycine buffer..
with or without methanol
wth or without SDS?
any differences? for what kind of proteins? i have done all these.. cant c any difference?
also how do you define efficiency of the transfer? by staining the gel after transfer? do u get like 100% transfers? pls let me know!

their are numerous transfer buffers; we routinely use 48 mM Tris, 30 mM Glycin, 0,037 % (w/v) SDS, 20 % (v/v) methanol;
staining gel after transfer is a good way to check transfer efficiency
