protein determination in loading buffer - (Feb/05/2008 )

Hi,

I want to do a western blotting.
The protocol attached to my 1st ab recommends
cells be directly lysed in loading buffer containing 2%
SDS.
For some reason, I want to determine the concentration
by BCA assay before SDS-PAGE.
I omitted beta-ME because it interferes with BCA reaction,
but should BPB be omitted from the buffer also?

yes. the color will interfere with the determination.
