Proteomics - Determination of structure of Protein. (Dec/17/2007 )

unsure.gif Why are fluorescence maxima observed at longer wavelengths than absorbance maxima?? wacko.gif Plzzzz help me out!!!!!!

some of the energy was absorbed by protein. so the energy of fluorescence is lower than the energy of absorbance.
therefore, the fluorescence maxima wavelength is lower than the absorbance maxima wavelength.
normally, the wavelength of absorbance is 280 nm due to trp and tyr but the wavelength of emission is 320 to 340 nm due to trp or tyr.

Thanks leehigh...

Now I understand... it is due to vibrational energy which is ultimately dissipated as heat... right??

But still searching the answer for the question why do peaks in absorbance spectra becomes narrower when the temperature is lowered to 77 K.... wacko.gif
