how to define an appropriate protein reaction buffer? - (Dec/02/2007 )

i am working on a glycanase of rhizobia. i want test the activity of the protein. however, no published paper mentioned the reaction buffer. how can i prepare the appropriate buffer? thank you very much!

you need to know the pH, salt concentration, cofactors (eg-Ca, Mg, edta, atp, etc) and substrate and the concentrations that your enzyme requires.

you can get starting points from the literature (of other species if yours has not been determined).
