How well do proteins extract from gel slices if 50mM of ammonium bicarbonate is only used, but no organic solvent (i.e: 10% ACN) was used throughout the entire extraction procedure? I know it will depend on the type of protein being extracted, but how about in general?

Do this question mean that you have tried the expt without ACN, and it hasn't worked?
Frankly, I would doubt you'd get much extraction.
