Linker design - Design of linker to fusion two protein domains (Aug/02/2007 )

Has anyone designed linkers to fuse two active proteins that usually work as dimers? How should I construct a peptide library and actually clone that?

Hi everyone,

I would like to bump this because I am experiencing a similar problem. I am about to design my first fusion protein with multiple domains that should fold independently from each other. For that, I am now looking for suitable linkers...

There is this publication by Crasto et al. ["LINKER: a program to generate linker sequences for fusion proteins" Protein Engineering 2000;13(5):309-312] that presents a software that would design linkers for me, but the URL in the paper is no longer valid and I can't seem to reach any of the authors by e-mail.

Does anyone of you know how to design such linkers? Is there any publication that I have missed which compares different types of linkers for fusion proteins?
