I got the problem of picking too much of a colony which led to the presence of aspecific bands on my gel. I will try to use the dilution step. Thanks!

I use to pick my colonie, and either scratch it of on a masterplate or move it aroud in a tube containing LB media for growing bacteria befor a dip in the PCR tube. My PCR mix contains triton-X
