hi gyes could some body tell me what is the first step after transformation of competent cells i already got white colonies , do i have to streak these colonies on ampicillin plates again????
or plasmid mini prep followed by pcr using primers of the insert???

hi again today I've done plasmid miniprep but when i run mini gel to imy plasmid i've got nothing !!
is it possible i thought that in both ways i should have plasmid even if it is not recombinant my questhion is: this white colony is either a transformed one with my recombinan or mutation somebody please answer me

after mini prep did you saw any DNA pellet???

no plasmid frequently means no antibiotic added during culture.
check on that.
After, you may loose your plasmid DNA as T.reesei said previously
