Gel Loading- Equal Protein or equal volumes? - (Apr/09/2007 )
Hello!
Been thinking of the pros and cons of gel loading by equal amount of protein in tissue homogenate vs. loading equal volume of homogenate for all samples.
What are your opinions??

It depends what you want to see.
If want to know if a protein is phosphorylated, I load equal quantity of proteins.

if you fractionize a homogenate f.i. in a sucrose gradient, you may analyze volumes to demonstrate amount of protein per fraction;
as missele says, the aim decides what you do; give us a concrete example to discuss
