anyone had elute protein from PVDF and use it for mass spec? Thanks - (Mar/23/2007 )

Does anyone has eluted protein from PVDF and follow with mass spec?

Could you give me the protocol?
I had tried with silver stain band, although I saw a hugh band, but still could not get mass spec properly.
And also I have lots keratin contamination, do yoou have any suggestion that I could prevent it?

I´m not an expert but I think that to avoid keratin contamination you can cut your band under a laminar flow cabin (as a cell culture), use gloves (although I suposse you are already using them), avoid standing above the samples in order to prevent any hair or your own invisible cells from falling on the sample...
Also some mass spect services have defined known keratin peaks which are subtracted from your sample.
