Very huge vector for transformation - (Mar/18/2007 )

I got a 21 kb construct in pBINPLUS vector and I am not successful with my normal competent cell. Could you please suggest any good and cheap ultra competant cell which is suitable for my transformation. Thanks in advance.

unless your plasmid has tandem repeats in it, any company made competent cell will take up a 21kb plasmid.

21kb isn't big. The home made competent cells my lab uses will take 21kb quite happily.

what i canrecommend is recA- cells and a heat shock transformation rather than electroporation.

I'm always more succesfull with electroporation if the plasmid is over 10 kb.

I just transformed 33 kb plasmid in DH5 cells and it worked
