serine protease - (Mar/15/2007 )

hi all,
i am new to this forum.
I am trying to express an insect gut trypsin in pichia pastoris, my construct has his and myc tags in pPICZalpha vector, however, the protein is not being secreted to the medium, i could see some immunoreactivity wehn i tried with cell lysate protein preperation. has anybody any idea, why it might not be secreted into the medium?

Does your plasmid direct proteins to the medium? Or do you have an export signal?
