Hi everyone,
Can anyone tell me is there any difference in using TBS or TBS w/Tween 20 during the blocking step of western blotting?
Which one is recommended to mix up with non-fat milk?

Tween 20 serves as a non-protein blocking agent, you can mix it with non-fat dry milk or leave it; washing steps after blocking are recommended to perform with TTBS

we use TBS with tween for preparing our blocking solution.

we block without tween. we use tween in our antibody and wash solutions. it helps reduce non-specific binding.

It's useless to put tween in blocking, however it won't be bad. So, do as it is more convenient for you.
