How many methods can demenstrate to endogenous protein interactio? - (Feb/12/2007 )
Hi, everyone:
I am doing IP to demonstrate two proein interaction. I have successfully showed that ectopic protein A and protein B( by transiently express the two proteins in cells) can interact with each other. To get more covinced result, I need to demonstrate endogenous interaction. However, the problem is protein A is about the same size of heavy chain, and protein B is at size of light chain. Is any way to overcome this problem in IP or is there any other method to demonstrate two endogous protein interaction other than IP?

there is this exciting new technique called P-LISA: Proximity Ligation and Specific Amplification which offers you the possibility to monitor endogenous protein-protein interaction in situ. I think it is tricky but the guys at Rudbeck Institute are really kind.

There is FRET, as well as PCA.
