confusing shape of survivin protein band - (Jan/22/2007 )

Dear all
I got confusing shape of survivin protein band on western blot, all bands shown in the attached image represent survivin from the same cell line analysed on the same gel and blotted on the same PVDF membrane. the only difference is that the 3 bands on the left hand side were obtained from the first replicate of the experiment and survivin band appear as a single band while the 3 bands on the right hand side were obtained from the second replicate of the experiment and appear as 2 bands!!
I do not know what is the reason for this difference, does anyone has a suggestion about this?

if the new band is lower then proteolysis?

if the new band is larger it could be aggregation or binding of another protein, but can not tell without knowing the weights of the bands and if there is disappearance of another band.

Actually the weight of all bands appearing on this blot is 16.5 KD. there is no disappearance of another band on the blot. this problem is happening with me frequently only when I analyze survivin. what do you suggest to overcome?

i'm not familiar with survivin. does it bind anything (oxygen, co2, metals)? does it get phosphorylated?

it looks as though you may be getting 2 species, one with and one without something attached. and it is occurring spontaneously, over time.

by the way, it looks like you may have two species in your earlier samples, but the balance or separation is different.
