cloning size - plasmid (Jan/08/2007 )

how big a vector can be made using common techniques. Can a vector of 25 kb or 50 kb be made, or only 10-12 kb? I know that BACs can be larger. Is it difficult to make BAC vectors?

i posted here my protocol to prepare bacmids from bacterias. It's not that difficult, just take care when pipetting and do not vortex
