will linearizing plasmid prolong transient transfection ? - (Nov/14/2006 )

hello all,

Has anyone got any experience of improving transfection or prolonging gene expression in transient transfections by linearizing plasmid ? will it even change the transfection charactersistics at all ? (I use nucleofection in human T-cells)

I'm asking this because I'm making stable transfections & I notice that cells transfected with linear plasmid seem (to my inexperienced eyes anyway) to be in better shape over the first few days in culture compared to those transfected with circular DNA.

Linearized plasmids increase the frequency of stable transfections, but reduce the initial transfection efficiency.
