thanks so much for those extra tips--yeah, my boss and postdoc in the lab also said BHI+Cm+Erm should be fine-so I will use such plate in my next trial....

As for concentrating DNA, I used Zymo column(a brand name), it can concentrate the DNA into 6ul of volume--so far, I found it better compared with the Qiagen minielute columns.....
