just got the results of WB. no cleaved band where i thought . so simply there is no cleavage. all i can think of is: can GST interfere with cleavage, since it is at the C-terminus where the cleavage is supposed to happen?

yes, it is possible that the bead is physically preventing the protease from acting on your protein.
