Urea in SDS-PAGE sample - (Sep/20/2006 )
Hi everybody. I´m planning to use an urea-based protocol for purification of a His-tagged protein in denaturing conditions. The protocol recommends adittion of urea (8M throughout all the purification) in the elution buffer as well.
My question is: is such a high urea concentration compatible with a standard SDS-PAGE electrophoresis? They do not mention any dyalisis step or anything.

Yes it is compatible.

(just to know : sometime guanidine hydrochloride is better for solubilization of some proteins : but this is not compatible with SDS-PAGE. you have to add a precipitation step to get rid of guanidine.)
