IEF for pI 9.4 protein - ? (Jul/18/2006 )

Is it possible to do an IEF for a basic pI protein say, pI 9.4? I have most literature saying cathodic drift will hamper this.. any one any suggestions? Thanks!

you can use 8-10.5 ampholytes either for just that range or to enhance a wide range separation. this should help avoid distortion due to cathode drift.

you might also try using immobilines or precast immobiline gels. the immobilized pH gradient will resist cathode drift.
