plasmids smaller than empty vector - using expensive cells - still not working (Jul/14/2006 )

hi all


experienced cloner seeking help!!!!

2.5 kb insert + 6.5kb vector (pFLOX) in XL10gold cells........i think i'm getting some crazy recombination or something because my plasmids are all way way smaller than empty (screened over 100 colonies from various ligations, all exactly the same size)


any suggestions? i've heard that 25C instead of 37C might help????

Do u have the possibility to get cells which prevent recombination like SURE cells or Stbl3 cells.
This might help.
