depends on whether you want protein from cellular components like erythrocytes or from plasma.....

in plasma you will have to precipitate whole protein...

in cellular parts you disrupt cells diluted in lysis buffer...

Thanks. I would like to get ride of the RBC with red cell lysis buffer. Then would like to use the cell lysis buffer. Hope it helps.

Hang on. Do you really want all of the RBC components in the purification, because that's what's going to happen if you use red cell lysis buffer on whole blood? (Unless I have completely misread your last message, in which case, I apologise)
If you want the plasma proteins, I'd suggest you simply spin down the cells and pour off the plasma, then treat it. If you want cellular proteins, you can do a density spin, to separate the white blood cells, RBC and plasma (I think that platelets are also removed...), then you can use whatever protocol you like as appropriate.

I will do a ficoll seperation. Then treat the RBC, WBC and plasma seperately.
