hello
i want to ligate a dna fragment (289pb) to do a long fragment
I have the fragment with blunt ends and too with cohesive ends (SalI/XhoI)
which volume and unit of T4 dna ligase must i do ?

I do ligation in a total volume of 20ul, but there r people who do it in 10ul.
I use 1ul of T4 ligase (NEB).
