Does the T4 ligase works perfectly well in the presence of TAE and EtBr? - (May/26/2006 )

I would like to know if T4 ligase works well in the presence of TAE and EtBr.

Using the “GenElute™ Agarose Spin Columns” (SIGMA), you can “purify” DNA form a slice of agarose gel. But you remove just the agarosa, and DNA is still eluted in TAE with EtBr…. The manual says DNA can be used in ligation.
My question is ….. Does the T4 ligase works perfectly well in those conditions??

Thanks!!

well i did lots of elutions using Qiagen with good results upto expression vectors so to my knowledge TAE and etBr creates no problem.
go ahead and good luck
