expression in Rosetta pLysS - how to get a good expression level?? (May/25/2006 )

Hi,
I'm trying to express a toxic gene in E. coli. So I have to use pLysS strain to prevent basal expression and culture dead but the protein expression was very poor.
Has someone any tip to improve it??
I need almost 10mg of my protein urgently! Help me please!!

Is it a eukaryotic or membrane protein?

What exactly are you trying to express?
