enzyme purification - (Apr/12/2006 )

Hi, I need to isolate an enzyme from C elegans. I have been searching in the net and in books and i really found many ways to do it. So i got a little confused. Is it there someone that had done this? I really need some advice about wich is the best method.

A lot of enzymes need their own procedure for cleaning. Steps like centrifugation, precipitation, different columns (SCX, gel filtration), pH, conductivity...
