as up, any software or website can calculate the protein charge?
I want to use separose column, so need to know wheter m y protein is negative or positive charge.

Go to Expasy website you will find many tools for protein including the PI determination

ExPASy
There you can paste your sequence, get the pI and you can decide, weather you use a more acidic puffer below the pI (protein is protonated) or you want a alkalic buffer for negative charged proteins.

Just to be different, you can also use PepStats, part of the Emboss package. See here.
