Problem of protein expressio in different medium - help! (Jan/10/2006 )
my protein used to have good expression in both E.coli C41 and BL21 (in LB and M9ZB)
however, when i change the medium to SOC, the cells stop growing and gives me a very small cell
pellet (only 1/4 to the normal yield)
But my labmates also use SOC for expression (different protein from mine), it seems that there is
no problem to their proteins)
So i think it should not be the problem of the competent cells. Can anyone had ever faced this
problem?
i am really eager to figure out what's going wrong with my expression!~~

i think u must have prepared your competent cells as well transformation in SOC medium only, if not then this is the only problem. u should do all the steps from starting in SOC only.
