attach adaptors or PCR the restriction enzyme sites? - which one would you suggest and why (Dec/06/2005 )

hello everyone...smile.gif

i want to cut a piece from the plasmid with my cDNA and transform it into GST vector....so i can either cut it and then amplify by PCR the restriction enzyme sites and then ligate or cut and buy adaptors and then ligate.....why would you prefer the one over the other? thanx a lot

definitely pcr adding site. you chose the sites and control the DNA amplified. You don't have such possibilities with a restriction/adaptators.
Finally pcris quicker.

You can control directionality if you design two different restriction sites into the primers. You can't do that with the adaptors.

Definitely do what you need to do with the primers.
