What media would promote the best E.coli growth at a low cost for fermentation?

-michaelw-

anyone?
BHI i know will make E.coli grow very very fast, but is there something cheaper?

-michaelw-

You could try minimal media as it doesn't really contain any other than glucose and salts (both cheap). If you feed slowly you should be able to get to a high cell density using a fermentor.

DNA FAQ

hiya
the fermentation is a batch process, and will run over an 8 hour period maximum..

Well you will need to use either a large inoculum or very expensive rich media. Eight hours is a very short time for a fermentor culture. Why so short?

I dont know, its not up to me
We start with a 1ml culture of E. coli in glycerol stock, then 200ml shakeflask, then 10L fermenter.
what would the best media be? just tryptone + phosphates or something?

-michaelw-

If you are only growing 10L just use on of the rich media like super broth of terrific broth.

it has to be cheap though..
we dont have super or terrific broth and we have a limit on how much we can spend
we have basic ingredients like casamino acids, bactopeptone, soypeptone, phytone, fructose, glucose, mgcl2, kcl, phosphates, nacl, bhi, etc
bhi is what id choose but its too expensive..

You can just make it up these broth's from Sambrook. Do you have yeast extract?

Have a look here for what you need.

neat thanks
yes i do have all those ingredients

but im not looking to isolate plasmid dna or anything.. i have to get a high yield in a short amount of time. i think the recipes they give there is for slow growth

"While cells grown in Terrific Broth grow slower than in LB the final cell density and plasmid yields are 2-4 fold higher. It is recommended that cells grown in TB are allowed to grow for 24 hours to achieve high densities"

Unfortunalty I have 8 hours for the entire fermentation..

any media modifications that you can suggest I make to 'terrific broth'?
