Problem with BsaB1 digestion - (Oct/15/2005 )

dose any person use BsaB1. last two week I try many times to use this enzyme to cut pires2egfp. but it is not work.
could you give me some advice

I've used it -- not often, but successfully. What problems are you having?

In looking at the enzyme just now, two things jump out -- the digests must be done at 50°C, and the enzyme is sensitive to dam methylation. See here.
