Human collagen type I molecular weight - (Aug/10/2009 )
Hello
I am trying to look for molecular weight for human collagen type I ...but i can`t find
please any help
(i want to do western blotting for collagen type I )
but i dont know where collagen bands should appear

monomeric collagen type I consists of alpha-1 and alpha-2 chains.
Alpha-1 is 139 kDa and alpha-2 is 129kDa.
As collagen type I is a triple helix consisting of one alpha-2 chain and two alpha-1 chains, you may also get bands of the dimeric (~270 kDa) and the trimeric form (~400 kDa).
Remember that those chains are cross-linked and can't be broken by typical sample denaturation for SDS-PAGE.

so what is the preferable conditions to run sds-page and WB in your experience ?

sorry, I don't work on this - the only experience I have is from a lab course a couple of years ago...
I guess we denatured the samples with guanidiniumchloride, which breaks the hydrogen bonds, but I don't have the exact protocol here... 
Maybe the others know?
