Best yeast expression system - (Jul/02/2009 )
Dear all
We want to start protein expression in yeast. Our proteins are human - so we want to go for glycosilation, and mostly unsoluble in E.coli - so we hope to get them soluble.
As I have not done this before could you please tell me your experiences in this field ?
Which yeast system is best for my task (we also want to produce good amounts of protein) ?
I read something about the K.lactis system from New England Biolabs, this sounds very convenient - is somebody working with this ?

Hi pET,
we are currently using the K.lactis system and getting good results. It is simple and the passing from e.coli to yeast is simple (i.e. the cloning part), but I will say is that as there is no antibiotic you need to work very clean and efficiently, also we had some problems with the competent cells not selecting properly (now resolved). That being said NEB were VERY good with any problems/questions we raised.
Is it better than other yeast systems? I donīt know but it is a valid option, very cheap as no antibiotics are used and the expression levels are as good as any I have seen (personal observations).
I have no connection with NEB nor any company selling ANY commercial product.
