Storage of 8M Urea buffer? - I made it two weeks ago? (Jun/24/2009 )

Hello,

I made a some 8M Urea buffer two weeks ago and since then Ive had it at room temperature. I read somewhere that u used always make it fresh and/or store it at 4C? Is this true then I cant use my big bottle of buffer :)

I hope someone can tell me this isnt true hehe

Oh and my buffer consists of Urea, Tris and NaH2PO4 in case it makes a difference

What u heard is true and will affect denaturing purification (I am assuming that is what u plan to do with the buffer)- to what extent, can be judged by doing a mini-scale purification.

Not recommended to pre-make urea containing solutions- u can try salvaging what u have if it still works in ur application.

Urea in water releases isocyante, which will carbamylate yr proteins. Make fresh buffer. Read this:
